چکیده

Introduetion Apple proliferation (AP) is one of the most economically important phytoplasmal diseases in Europe. It seriously impairs fruit size, yield, and vigor of the trees. AP was first reported in northern Italy (Rui et al., 1950). In the following decades, the occurrence of AP was reported from most European countries. Due to its transmissibility by grafting, the disease was initially thought to be caused by a virus. Later, wall-less bacteria, then named mycop1asma-1ike organisms, were identified in symptomatic trees (Giannotti et al., 1968), shortly after this new group of plant pathogens was discovered (Doi et al., 1967). Restrietion fragment 1ength polymorphisms (RFLP) and sequence analysis of ribosomal DNA revealed that AP is induced bya distinct phytoplasma that is closely related to the pear dec1ine (PD) and European stone fruit yellows (ESFY) agents, the causes of two other major diseases of temperate fruit trees (Seemüller et al., 1998). Unlike most or all other phytoplasmas, which are transmitted by either leafhoppers (Cicadellidae) or planthoppers (Fulgoroidea), the three fruit tree phytoplasmas are mainly or only spread by psyllids (Psyllidae) (Frisinghelli et al., 2000; Tedeschi et al., 2002). Recently, the AP phytoplasma was delineated taxonomically under the provisional status 'Candidatus' for uncultured bacteria as 'Candidatus Phytoplasma mali' (Seemüller and Schneider, 2004).

Apple Proliferation Phytoplasma Influences the Pattern of Plant Volatiles Emitted Depending on Pathogen Virulence

Apple proliferation (AP) and pear decline (PD) are the most severe diseases in pome fruit growing areas. AP-infected trees show typical symptoms such as witches’ broom, enlarged stipules, tasteless, and dwarf fruits. PD-infected pears show a progressive weakening, reduced terminal growth, smaller fruits, and die within weeks (quick decline) or years (slow decline). The diseases are caused by th...

Physical map of the chromosome of the apple proliferation phytoplasma.

A physical map of the apple proliferation phytoplasma strain AT chromosome was constructed from genomic DNA extracted from diseased tobacco plants. The map was generated with single and double digestions of the chromosome with BssHII, SmaI, MluI, and ApaI restriction endonucleases and resolving the fragments by pulsed-field gel electrophoresis. Partial digestion and Southern blot analysis were ...

‘Candidatus Phytoplasma mali’: the current situation of insect vectors in northwestern Italy

The two acknowledged vectors of ‘Candidatus Phytoplasma mali’ in north-western Italy, the psyllid Cacopsylla melanoneura and the leafhopper Fieberiella florii, have been compared to highlight their role in the apple proliferation epidemiology in relation to their infectivity, transmission efficiency and permanence in apple orchards and in wild vegetation, predominantly hawthorn formations. The ...

Phloem cytochemical modification and gene expression following the recovery of apple plants from apple proliferation disease.

Recovery of apple trees from apple proliferation was studied by combining ultrastructural, cytochemical, and gene expression analyses to possibly reveal changes linked to recovery-associated resistance. When compared with either healthy or visibly diseased plants, recovered apple trees showed abnormal callose and phloem-protein accumulation in their leaf phloem. Although cytochemical localizati...

Establishment of a new method for rapid and precise estimation of apple proliferation phytoplasma concentration in periwinkle.

Quantification of a plant pathogen is essential to study its population dynamic in various conditions and to relate symptom expression with pathogen concentration. Up to now, very few methods have been published to quantify phytoplasmas. So, the objective of this work was to establish a method able to quantify the Apple Proliferation (AP) phytoplasma populations in periwinkles. The present work...
